Maintenance of an Isolated Smooth Muscle Preparation In Vitro

Overview

This course would look at maintaning an isolated smooth muscle prepared in vitro, that is, outside the body. The Harvard Kymograph with stimulator is the equipment that enables the recording of contractions of muscles from the isolated tissue preparation. The video lessons highlight the conditions neccessary for the maintainance of a smooth muscle prepation.

 

Our objective is to investigate the effects of changes in oxygenation, glucose, temperature, ionic constituents and pH on the rabbit jejenum using isolated organ bath at 37 °C.

Introduction

 

In this laboratory procedure, The Harvard Kymograph with stimulator, when set up will be used to record the contractions of muscles from the isolated tissue preparation. In order to investigate the conditions neccessary for the maintainance of the preparation, suitable adjustment should be made. These video lessons demonstrates the setting up of the smooth muscle preparation, the adjustments on the Harvard Kymograph and its results. You would aslo observe some form of biological activity during the the demostration of the procedure.
